Immunomodulatory Activity of Kedondong Leaf Ethanol Extract (Spondias Pinnata) (L.F) Kurz) against the Proliferation of Balb/C Mice Lymphocyte Cells Along with Flavonoid Content Tests

Continuous exposure to pathogenic foreign bodies can lower the immune system, so the body needs immunomodulatory compounds containing flavonoids. Flavonoids can increase lymphocyte cell proliferation. This study aims to determine the immunomodulatory activity of kedondong leaf ethanol extract against the proliferation of balb/c strain mice lymphocyte cells induced by hepatitis B vaccine along with flavonoid content tests Kedondong leaf extract was extracted by maceration with 96% ethanol The extract was administered on a p.o basis in the treatment group of doses 5, 10 and 20% positive control of levamisol l0.45 mg/20gram BB, control with CMC-Na vaccine 0.5% and normal control for 46 days. Lymphocyte cells were isolated from the spleen and tested for lymphocyte cell proliferation using MTT Assay. Identify flavonoid extract compounds with Mg and HCl reagents. Optical Density (OD) results analyzed by Kruskall-Wallis Test statistics followed by Mann-Whitney with a 95% confidence level. Meanwhile, the identification of chemical content is compared to the reference library. The results showed that kedondong leaf ethanol extract has activity against increasing lymphocyte cell proliferation at a concentration of 10% and contains flavonoid compounds. Kedondong leaf ethanol extract has immunomodulatory activity and contains flavonoid compounds.

Abstrak
Paparan benda asing yang bersifat patogen secara terus menerus dapat menurunkan sistem imun, sehingga tubuh membutuhkan senyawa imunomodulator yang mengandung flavonoid. Flavonoid dapat meningkatkan proliferasi sel limfosit. Penelitian ini bertujuan untuk mengetahui aktivitas imunmodulator ekstrak etanol daun kedondong terhadap proliferasi sel limfosit mencit galur balb/c yang diinduksi vaksin hepaitis B beserta uji kandungan flavonoid. Ekstrak daun kedondong diekstraksi secara maserasi dengan etanol 96%. Ekstrak diberikan secara p.o pada kelompok perlakuan dosis 5, 10 dan 20% kontrol positif levamisol l0,45 mg/20gram BB, kontrol dengan vaksin CMC-Na 0,5% dan kontrol normal selama 46 hari. Sel limfosit diisolasi dari limpa dan diuji proliferasi sel limfosit menggunakan MTT Assay. Identifikasi senyawa flavonoid ekstrak dengan pereaksi Mg dan HCl. Hasil Optical Density (OD) dianalisis statistik Kruskall-Wallis Test dilanjutkan Mann-Whitney taraf kepercayaan 95%. Sedangkan identifikasi kandungan kimia dibandingkan dengan pustaka acuan. Hasil analisis menunjukkan bahwa ekstrak etanol daun kedondong memiliki aktivitas terhadap peningkatan proliferasi sel limfosit pada konsentrasi 10% dan mengandung senyawa flavonoid. Ekstrak etanol daun kedondong memiliki aktivitas imunomodulator dan mengandung senyawa flavonoid.
